Optimization of Enzymatic Extraction of Peanut Protein by Response Surface Methodology

摘要
Four kinds of proteases including Alcalase 2.4L,2709 alkline protease, AS1398 neutral protease and Protex 6L were used to extract peanut protein, respectively. 2709 alkline protease showed the highest extraction efficacy, followed by Protex 6L. Response surface methodology (RSM) was used to study the effects of 2709 alkline protease on extraction of peanut protein. The highest protein yield was 94.3% under the optimal conditions (solid-to-water ratio 1:4, alkaline pH 9.0, enzyme amount 3.75g (in 60g of peanut seeds) and hydrolysis time 1.5 h. By analysis of the experimental results, a suitable model was obtained for predicting the yield of protein (R-2 = 0.8182, p<0.01). The solid-to-water ratio highly significantly (p<0.01) affected the protein yield.
