The IL-23/Th77 pathway has been recently linked to the pathogenesis of autoimmune disorders, and a number of novel therapeutic agents that target this pathway are being developed. Brodalumab (AMG-827) is a human anti-IL-77-receptor A (IL-17RA) monoclonal antibody being evaluated in a number of early clinical studies for the treatment of inflammatory and autoimmune indications, including plaque psoriasis, psoriatic arthritis, rheumatoid arthritis, asthma and Crohn's disease. Brodalumab has demonstrated a high level of response, with a rapid onset of action, and has recently advanced to phase III trials for the treatment of moderate to severe plaque psoriasis. However, development of brodalumab for the treatment of rheumatoid arthritis and Crohn's disease has been discontinued due to undesired results from the early clinical trials in these indications.
