Effects of Hg2+ on Luciferase mRNA Expression and Bioluminescence of Photobacterium leiognathi

摘要
Objectives The aim of this study was to investigate the effects of Hg2+ on the mRNA expression of luciferase genes, as well as on the emission activity of the bioluminescence enzyme system. Methods Hg2+ was added into culture medium of Photobacterium leiognathi to detecte gene expression of luciferase by semi-quantitative RT-PCR, and was mixed with extracted enzymes to detecte the inhibitory effects of Hg2+ on the bioluminescence system. Results Expression of luciferase mRNA in the experimental group (0.83 <= 0.13) was significantly higher (P<0.01) than that in the control group (0.45 +/- 0.01) at the beginning of the log phase. The toxic effect on the bioluminescence system of enzymes increased proportionately with the concentration of Hg2+. Conclusion Hg2+ in low concentration (<= 0.5mg/L) can induce the expression of lux genes during the early stages of log phase, but it inhibits the intensity of the bioluminescence system of enzymes.
