Direct resolution of ibuprofen enantiomers by reversed-phase high performance liquid chromatography with amide derivative as chiral stationary phase

摘要
By using reversed-phase high performance liquid chromatography with amide derivative [(R)-1-naphthylglycine & 3,5-dinitrobenzoic acid] as chiral stationary phase (CSP), the direct resolution of racemic ibuprofen was investigated. The resolution was optimized by varying the concentration of methanol, and the pH in the mobile phase. The organic modifier was also altered by substituting acetonitrile and tetrahydrofuran (THF) for methanol. The optimized mobile phase was methanol/H2O (70/30, V/V), containing 0.01 mol/L NH4Ac and pH 5.9 (with HAc). Although the racemic ibuprofen couldn't been direct resolution by HPLC with urea derivative [(S)-Valine& (R)-1-(alpha-naphthyl)ethylamine] as the CSP, the resolutions of ibuprofen uith these two CSPs are agreement with the three-point rule.
