摘要
The temperature effect on DNA fragment separation in hydroxyethyl cellulose (HEC) media was investigated. The migration time was shorter, but the separation efficiency was improved at elevated temperature for Phi X174/HaeIII separation. The closed 271/281 base pair was successful separated at 55 degrees C in both 0.7 and 1.0% HEC. The activation energy tendency for migration of different fragments showed that there were significant changes for fragments in the 200-400 base range in 1.0% HEC and in the 400-600 base range in 0.7% HEC.
