Cucumber mosaic virus in globe artichoke in Turkey

A survey was conducted to identify viruses infecting globe artichoke (Cynara cardunculus L. subsp. scolymus (L.) Hayek) in the most important globe artichoke-producing areas of Turkey. A total of 120 globe artichoke samples were collected from three provinces of the Aegean region in western Turkey. Samples were tested by DAS-ELISA, RT-PCR and mechanical inoculation onto indicator plants. Cucumber mosaic virus (CMV) was detected in five of the samples tested. Tomato spotted wilt virus (TSWV) and Tobacco mosaic virus (TMV) were not detected in the remaining 115 samples. In RT-PCR, amplicons of the expected size (about 540 bp) were obtained from symptomatic plants that were CMV-positive in DAS-ELISA. The sequenced 5 PCR products covering the partial CP gene revealed 99% identities at the nucleotide level with those CMV isolates. When inoculated with sap from symptomatic artichoke leaves, selected test plants developed the symptoms that are typical of CMV infection. This is the first record of CMV infecting globe artichoke in Turkey.
