The Use of Ovule Embryo Culture for Obtaining Triploid Plants from Cross between Diploid and Tetraploid Grapes

摘要
Triploid grape breeding is one of the effective approaches to obtain new seedless cultivars but it is difficult to get triploid progenies by conventional methods. The present work was carried out to determine the efficiency of ovule embryo culture for obtaining triploid plants from cross between diploid and tetraploid grapes. The different stages of embryo development and abortion were investigated. The effects of sampling period, basal media, and beak-cut treatment in the crosses between diploid 'Muscat Hamburg' (V. vinifera) and tetraploid 'Kyoho' (V. vinifera x V. labrusca) grapes were studied. The optimum protocol consists in sampling ovules 71 days after pollination and using B5 medium supplemented with IAA 0.1 mg/L and IBA 0.1 mg/L. The percentage of germination and plantlet formation are 51.0% and 36.0%, respectively. The ploid level of plants was determined by flow cytometry.
