Sex chromosome abnormalities in spermatozoa of fertile and infertile men detected by FISH

This research was carried out to analize the numerical aberrations of sex chromosome which cause genetic disorders in human sperm. Sperms were collected from 10 nomal males and 10 oligospermic males. A total 46,648 sperm nuclei of normal males and 42,766 sperm nuclei of oligospermic males were analyzed by fluorescene in situ hybridization (FISH) with dual-color specific probes. This technique, using only probes (Vysis) for sex chromosome and not for autosomoes, does not discriminate the differences between hyperploid and diploid sperm. The frequency of chromosome XY aberrations was significantly higher in oligospermic males than in normal males (0.001 < P < 0.01). Also, the frequency of chromosome YY is significantly higher in oligospermic males than in normal males (0.01 < P < 0.05) These results suggest the abnormality of sex chromosome at meiosis I or II might cause infertilization and some genetic disorder in human.
