A local application of an antioxidant for Cisplatin-ototoxicity

We investigated the effects of a locally applied antioxidant, trolox on CDDP (Cisplatin)-ototoxicity in guinea pigs. Drugs were put on round windows after opening the bulla via a post-auricular approach. The combined treatment of animals with trolox distinctly suppressed the CDDP-induced ototoxic side effects. These were: elevations of ABR thresholds at 4, 8 and 16 kHz; and substantial outer hair cell losses in the basal and second turns. Renal function was not impaired in each group. These results suggest that locally applied trolox provides a protective effect on CDDP-ototoxicity via the suppression of production of reactive oxygen species.
