Cytotoxic Effect of Doxorubicin on Human Multiple Myeloma Cells In Vitro

摘要
Cytotoxic effect of doxorubicin in human multiple myeloma cell cultures RPMI 1640, RPMI 8226, and IM-9 was studied. The obtained data were compared with mRNA expression of MDR1, MRP1, BCRP, LRP genes responsible for the development of multiple drug resistance. IM-9 cells that differed from the other two cultures by the expression of surface differentiation markers and by mRNA expression of MDR1, BCRP, and LRP were most sensitive to doxorubicin. All cells expressed mRNA for only A-isoform of insulin receptor (IRA), while B-isoform (IRB) was not expressed. Insulin in a concentration of 5 mu g/ml had no effect on the cytotoxic effect of doxorubicin in the studied cells.
