摘要
An efficient and large scale in vitro clonal propagation of valuable medicinal plant by enhanced shoot crown explant proliferation has been developed by us. Crown explants from Chlorophytum borivilianum initially treated with Bavistin 0.1% and 0.1% Endosulphan and 0.05% Fluconazole were cultured in solidified modified Murashige and skoog (MS) media, each with different hormonal combination for the establishment of cultures, multiplication and rooting of plants. Several plant growth regulators BAP(6 Benzyl aminopurine), Kinetine (Kn), IAA (Indole Acetic acid), NAA(alpha-Naphthylacetic acid) alone or in combination were tested for their capacity to induce shoot from shoot crown explants. For establishment of the shoot crown explants 2 mg/l Kinetin and I mg/l IBA were necessary. Average 73 shoots were generated when established cultures were transferred to modified MS medium containing 3 mg/l Kinetine and 2 mg/l BAP. Rooting was initiated in the half strength modified MS basal medium containing 30 gm/l sucrose(w/v) supplemented with 1 mg/l IBA (Indole buteyric acid). Strong rooted plantlets obtained after 3-4 weeks growth were transferred to soil-sand vermicompost (1:1:1 v/v) with more than 95% success in hardening. High multiplication efficiency, phenotypic stability and yield ensure the efficacy of the protocol developed for the production of medicinal herb safed musli.
