Characterization and differentiation of cervical cancer cell lines using ATR-FTIR spectroscopy and multivariate data analysis

ABS T R A C T Vibrational analysis has become a powerf u l tool for unraveling differences during malignant transformation. Still , the interpretation of data might be complicated due to the amount of information generated by a l l chemical bonds present in this type of sample. Therefore, applying chemometric analysis and mathematical processes to the spectra helps understand sample differences. In this work, attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectroscopy was employed for spectral acquisition, and principal component analysis (PCA) was used for chemometric analysis to differentiate prima r y fibroblasts from cervical cancer cel l lines, HPV-16 positive SiHa, HPV-18 positive HeLa , and HPV negative C33A. PCA model was constructed using the 970 - 1150 cm � 1 range. The model exhibited clear segregation according to the cel l type. Finally , spectral deconvo-lution (band curve-fitting) in a peak 1080 cm � 1 reveals contributions from 1035, 1051, 1080, and 1120 cm � 1 peaks related to glycogen, DNA/RNA , and lactate. Comparing these results to the current understanding of changes in these molecules confirm that the study of vibrations is sensitive enough to propose new, more sen-sitive, and rapid methods for this type of disease.
