Flavonoid-Rich Extract of Portulaca grandiflora Hook. Attenuates xidative Stress, Biochemical Changes and Vascular Dysfunction in Atherogenic Model

Portulaca grandiflora, a flavonoid-rich plant in our environment, has many medical uses. The whole plant has anti-oxidant, anti-diabetic, and diabetic nephropathy-protecting effects. Methanol extract exhibits a higher percentage yield than ethanol and acetone, and the phytoconstituents, such as flavonoids and polyphenols, have strong antioxidant activity. Therefore, a study was conducted to evaluate the efficacy of a flavonoid-rich methanol extract fraction of the aerial part of Portulaca grandiflora Hook. (PGMEF) against HFD-induced hyperlipidemia, hemodynamic change, antioxidant levels, and vascular dysfunction in rats. All tests were carried out on 200 +/- 20 g albino rats that were randomly divided into five groups Blood samples showed biochemical, hemodynamic, and histological changes on day 29. PGMEF 200 and 400 mg/kg stabilised hemodynamic parameters (HR, SBP, DBP, MBP), produced vascular relaxation, and changed biochemical markers (reducing MPO and raising SOD levels) and disease biomarkers (cholesterol, TC, TG, HDL, LDL, VLDL, AI, and CRI). According to the aorta staining study, standard and preventive group rats had less myocardial injury-induced damage and necrosis; infarct size was also significantly reduced, and myocardial structural changes were significantly recovered in PGMEF-treated groups. The changes in body weight, lipid profile, antioxidant level, and cardiac tissue histology indicate that the 28-day PGMEF treatment restored the body's natural state. PGMEF may be used to treat vascular dysfunction and cardiovascular risk, according to current studies.
