摘要
A fresh mixed-ligand Zn(II) coordination polymer, namely, [Zn-2(tbta)(phen)(2)(OH)](n) (1), in which H(3)tbta is 1-(triazol-1-yl)-2,4,6-benzene tricarboxylic acid and phen is 1,10-phenanthroline, was successfully synthesized through the reaction between H(3)tbta, a dual-functional ligand, and Zn(NO3)(2)center dot 6H(2)O in the presence of the N-donor co-ligand phen. Its anti-cervical cancer activity was evaluated via Cell Counting Kit-8 (CCK-8) assay. Furthermore, they were found to act as tumor suppressor genes that promote tumor cell apoptosis. Finally, their clinical pathological features were observed to be closely related to poor prognosis. miR-5571 was predicted as the upregulator of murine double minute 2 (MDM2) in cervical cancer cells. After transfecting miR-5571, the accumulation of intracellular reactive oxygen species and cell apoptosis in cervical cancer cells were detected and determined using 2,7-Dichlorodihydrofluorescein diacetate (DCFH-DA) and Annexin V-FITC/PI. The direction binding of miR-5571 and MDM2 was proved via luciferase activity detection. This research proved the important role of miR-5571 may help in the further development of better and effective therapeutic approaches for cervical cancer treatment, and the Zn(II) complex could be the excellent candidate for the cervical cancer therapy.
