Development of a high-performance thin-layer chromatography method for the quantification of mangiferin in different extracts of Mangifera indica L. and its antioxidant activity

摘要
A high-performance thin-layer chromatography (HPTLC) method was developed with selectivity, simplicity and sensitivity in mind for the purpose of identifying and quantitatively estimating mangiferin in various extracts of Mangifera indica. A solvent mixture comprising acetone-chloroform-formic acid-water (2:10:0.5:1.0, V/V) was utilised as the developing solvent to achieve efficient band separation. Densitometric analysis was conducted at a wavelength of 350 nm. The range for linearity was determined to be between 0.2 and 1.0 & mu;g per spot. During analysis, it was observed that the ethanolic extract of M. indica exhibited a higher concentration of mangiferin in comparison with the dimethylformamide (DMF), acetone and dimethyl sulfoxide (DMSO) extracts. The methodology underwent additional validation to ensure its accuracy, robustness, linearity, specificity and precision to a linear relationship. Additionally, the antioxidant efficacy of M. indica extracts was evaluated through the utilisation of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. The ethanolic extract exhibited the most substantial antioxidant potential with a percentage inhibition of 70.32%.
