Metabolites analysis and new bioactive pigments from a Monascus purpureus strain with low citrinin

摘要
A Monascus purpureus strain with high mycelial biomass and no detected citrinin in liquid fermentation was selected from 20 strains. HPLC-DAD-HRMS analysis revealed that the EtOAc extract of the fermented mash contained 2 unknown compounds and 11 known metabolites of Monascus fungi. Isolations with preparative and semi-preparative HPLC resulted in four brownish red pigments. The two unknown pigments were identified with HRMS, CD, ECD, 1D and 2D NMR, and designated as monascuskaolin A (3) and B (8). The two known com-pounds were verified with 13C NMR as monascusazaphilone C and monasfluol B. Result of bioactivity tests showed that the two new pigments were able to significantly reduce the transcription levels of IL-1 beta, TNF-alpha, and TGF-beta, and significantly promote the proliferation of 293 T cells, suggesting that they have anti-inflammatory function and may be good for human kidney. Metabolites studies suggested that M. purpureus RCEF7056 is a safe strain with functional food utilization potential.
