Synthesis and Characterization of Linear Polyglycidol Derivatives: Surface Activity and Potential for Advanced Doxorubicin Delivery Application

摘要
In this study, a series of glutathione-responsive (GSH-responsive) amphiphilic comb polymers LPGE-SS-n were synthesized based on linear polyglycidol (LPG). The corresponding structure-property curves were established through a systematic evaluation of their surface properties, drug solubility enhancement, and drug loading ability. These analyses revealed a close relationship between the physical characteristics and the length of the comb-like side chains. By using LPG as the backbone, multisite functionalization was accomplished compared to conventional PEG-based materials, resulting in significantly enhanced surface activities. Furthermore, the doxorubicin@LPGE-SS-2 micelle demonstrated selective and enhanced antitumor activity in response to high GSH concentrations, making it a potent candidate as an efficient drug delivery material. Therefore, our study offered a valuable reference for their potential applications in chemical and pharmaceutical industries.
