摘要
We combined the CRISPR-Cas13a system with CMC chemical labeling, developing an approach that enables precise identification of pseudouridine (psi) sites at specific loci within ribosomal RNA (rRNA), messenger RNA (mRNA) and small nuclear RNAs (snRNA). This method, with good efficiency and simplicity, detects psi sites through fluorescence measurement, providing a straightforward and fast validation for targeted psi sites of interest. A streamlined and efficient method enables precise detection of pseudouridine (psi) at specific loci, by leveraging CRISPR-Cas13a and CMC chemical labeling for immediate fluorescence analysis.
