Switching Nitrofurantoin Polymorphic Outcome in Solvent-Mediated Phase Transformation and Crystallization Using Solvent and Additives

摘要
We demonstrate that several additivesand solvents allow switchingthe polymorphic outcome of solvent-mediated phase transformation experimentsand crystallization of the antibacterial drug nitrofurantoin. Polymorph & beta; is obtained from most of the solvents, whereas theselection of alcohols as solvents or the use of crystallization additivesprovides the formation of polymorph & alpha;. We alsodemonstrate that this can be linked to reversed apparent relativesolubility of nitrofurantoin polymorphs in these solvents or in thepresence of the respective additives. We propose that this could becaused by different surface-additive and surface-solventinteractions formed by each of the nitrofurantoin polymorphs, whichwould change the relative surface energy of polymorphs. Several additives and solvent allow reversing the polymorphicoutcome of solvent-mediated phase transformation experiments and crystallization,as well as the apparent relative solubility of polymorphs of the antibacterialdrug nitrofurantoin.
