岩藻多糖对免疫低下小鼠模型免疫功能和肠道菌群的调节作用

摘要
目的:探究岩藻多糖(FUC)作为功能食品对免疫低下小鼠免疫功能及肠道菌群的调节作用。方法:40只雄性BABL/C小鼠随机分为4组,用环磷酰胺构建免疫低下小鼠模型,每天灌胃给予干预组高、低剂量的岩藻多糖,对照组、模型组每天给予等体积生理盐水。通过酶联免疫吸附试验、流式细胞术、16S rRNA基因高通量测序等方法,检测免疫低下小鼠的免疫功能和肠道菌群。结果:免疫低下模型小鼠机体免疫功能明显下降,小鼠脾脏组织和肠道组织损伤严重;经FUC干预后,小鼠脾脏组织、回肠组织形态破坏得到明显改善,脏器指数、血清中免疫球蛋白和细胞因子的质量浓度明显提高(P<0.05)。菌群分析表明,属水平上,与正常组比,模型组小鼠盲肠内容物中拟杆菌含量显著升高(P<0.05),乳酸杆菌、罗氏菌含量显著降低(P<0.05);经FUC干预后,免疫低下小鼠肠道中失调的菌群得到良好的恢复;FUC干预的各组小鼠,肠道中菌群结构与正常组趋近。结论:FUC可有效提高免疫低下小鼠的免疫功能,并能有效缓解环磷酰胺造成的肠黏膜损伤,其作用机制可能与FUC对肠道菌群的调节作用有关。
