四氢嘧啶对人皮肤成纤维细胞衰老的影响

摘要
目的 探究四氢嘧啶(ectoine)对人皮肤成纤维细胞(HSFs)是否具有减缓衰老的功效及其具体机制。方法 采用不同浓度四氢嘧啶作用于HSFs 24 h, MTT法检测细胞活性,β-半乳糖苷酶(SA-β-gal)染色观察细胞衰老情况,流式细胞术检测细胞调亡和活性氧(ROS)的水平,qPCR检测p16、p21、p53 mRNA的表达水平。结果 MTT结果显示,与对照组相比,各四氢嘧啶处理组对细胞无毒性作用,随着四氢嘧啶浓度增加,HSFs细胞凋亡率、ROS水平以及p16、p21、p53 mRNA表达水平均显著降低(P<0.05)。结论 四氢嘧啶可通过降低细胞凋亡率、抑制ROS活性和减少p16、p21、p53 mRNA的表达来减缓HSFs衰老,这将为今后开发以四氢嘧啶作为主要成分针对HSFs的抗衰老产品提供一定的理论依据。
