和肤止痒方对AEW诱导的慢性瘙痒小鼠皮肤屏障功能及AQP3、Caspase-14蛋白表达的影响

目的:探讨和肤止痒方对丙酮-乙醚-水溶液(AEW)诱导慢性瘙痒小鼠经表皮失水(TEWL)以及皮肤屏障功能的保护作用。方法:将42只BALB/c小鼠根据体质量按随机数表法分为空白组,模型组,和肤止痒方低、中、高剂量组和阳性对照组,每组7只。采用AEW诱导慢性瘙痒小鼠模型,观察和肤止痒方对模型小鼠止痒作用,测定TEWL、皮肤屏障强度,HE染色观察皮肤组织病理学改变,免疫组化法检测皮肤组织中水通道蛋白3(AQP3)和Caspase-14蛋白表达水平变化。结果:与空白组比较,模型组小鼠抓挠次数显著增加,皮肤屏障功能强度显著降低,TEWL显著升高,皮肤中AQP3、Caspase-14蛋白表达水平显著下降(P<0.01)。与模型组比较,和肤止痒方中、高剂量组小鼠的搔抓次数显著减少,皮肤屏障强度显著增加,TEWL显著降低,皮肤中AQP3、Caspase-14蛋白表达水平显著增加(P<0.05,P<0.01)。结论:和肤止痒方可能通过上调皮肤中AQP3、Caspase-14蛋白表达水平,进而减少TEWL,增强皮肤屏障功能,以达到防治慢性瘙痒的作用。
