miR-138-5p调控SIRT1抑制结肠癌细胞增殖及迁移的研究

摘要
目的:探讨miR-138-5p对结肠癌细胞增殖及迁移能力的影响及其作用机制。方法:RT-PCR检测结肠癌组织和细胞系中miR-138-5p的表达,分析其表达水平与结肠癌患者临床病理参数的相关性。建立miR-138-5p过表达的结肠癌细胞系,CCK-8法检测各组细胞增殖活性,Transwell法检测细胞迁移和侵袭能力。生物信息学预测miR-138-5p可能的靶基因,并采用双荧光素酶实验进行验证。Western blot法检测沉默信息调节因子1(SIRT1)蛋白的表达,进一步过表达SIRT1,观察细胞增殖、迁移和侵袭能力的变化。结果:miR-138-5p在结肠癌组织和SW480细胞系中表达均显著下调(均P<0.05),且其表达水平与患者的肿瘤浸润深度、TNM分期和淋巴结转移显著相关。过表达miR-138-5p可显著降低结肠癌细胞的增殖、迁移和侵袭能力(均P<0.05)。miR-138-5p可抑制SIRT1野生型质粒荧光素酶的活性(P<0.05),且过表达miR-138-5p可有效降低SIRT1 mRNA和蛋白的表达(均P<0.05)。过表达SIRT1可部分逆转miR-138-5p对SW480细胞增殖、迁移和侵袭能力的抑制作用(均P<0.05)。结论:miR-138-5p通过靶向作用SIRT1抑制结肠癌细胞的增殖、迁移和侵袭,并影响结肠癌患者的肿瘤进展程度。
