copine 8在胰腺癌增殖、侵袭转移和凋亡中的作用研究

摘要
目的探究copine 8在胰腺癌中的生物学功能。方法利用GEO数据库比较copine 8在胰腺癌组织和癌旁组织中的表达差异,利用The Cancer Genome Atlas (TCGA)胰腺癌患者组织RNA测序数据库分析其对总体生存率的影响,并对高copine 8表达组和低copine 8表达组的差异基因进行GO、KEGG富集分析和GSEA分析。利用siRNA干扰胰腺癌细胞系CFPAC-1和AsPC-1中copine 8的表达,CCK8实验和集落形成实验检测细胞增殖能力变化,Transwell实验检测细胞侵袭转移功能的变化,使用Annexin V-FITC和碘化丙啶分析凋亡细胞比例。结果 copine 8 mRNA和蛋白均在胰腺癌组织中高表达,在癌旁组织中低表达,且与患者生存预后相关(P=0.033)。富集分析发现copine 8在胰腺癌中会影响WNT、Hippo、mTOR和ERBB信号通路,可能通过影响Hippo通路中关键分子YAP1、STK3、STK4和SGMS1的表达从而影响胰腺癌的进展。干扰copine 8表达后,胰腺癌细胞增殖、侵袭和迁移能力显著降低,且凋亡细胞比例明显增加。结论 copine 8在胰腺癌增殖、侵袭转移和凋亡过程中有显著作用,有较强的促癌效应。
