:
本实验将苏云金芽孢杆菌的d一内毒紊基因导入水稻原生质体井得到了具有这种基因的转化水稻植株。所用的重组质粒pGY61和pGYCK63带有一个由35S启动子, d 内毒素蛋白基因,刀一葡萄着苷酸酶(GUS)报告基因和N。S终止子组成的嵌合基因。d 内毒素基因与GUS基因是融合基因, 而且GUS基因在d一内毒素基因的下游。水稻原生质体是从细胞悬浮系中分离出来, 用聚乙二醇(PEG)方法将带有d内毒素基因的质粒导入水稻原生质体。 经荧光法测定, 由这些原生质体产生的愈伤组织具有很高的/6r葡萄糖昔酸酶活性,说明J(I葡萄糖苷酸酶基因在水稻愈伤组织中得到表达, 表明d内毒素基因抬水稻愈伤组织中已被翻译(表达)。 另外, 从这些转化的愈伤组织中得到正常的绿色植株、 经分子杂交(Southem B]。t)证明d呐毒素基因已整合进水稻基因组中。
