PCR is a rapid method for amplifying a defined sequence from small amounts of genomic DNA or mRNA. There is a potent inhibitor of Taq polymerase in some samples of genomic DNA that blocks amplifying but is nevertheless a good substrate for restriction enzymes. The inhibition cannot be overcome by extracting with phenol and chloroform and
