COPD患者肺泡及外周血单核源性巨噬细胞Cofilin的表达与其吞噬烟曲霉功能的相关性

摘要
目的探讨慢性阻塞性肺疾病(COPD)患者肺泡及外周血单核源性巨噬细胞Cofilin的表达与其吞噬烟曲霉功能的相关性。方法选取2015年7月至2016年5月南方医科大学珠江医院呼吸内科门诊20名COPD患者,其中观察组1(2011版GOLD中A和B组),观察组2(2011版GOLD中C和D组),健康对照组10人。3组行支气管肺泡灌洗术、采取外周血,分别提取AM和MDM,检测各组的AM和MDM对烟曲霉孢子的吞噬能力。运用实时荧光定量(qRT-PCR)和免疫蛋白印迹实验(Western blot)检测3组Cofilin蛋白的表达,比较其差异。结果 3组人群MDM/AM吞噬烟曲霉孢子的菌落数分别为(17±3)个、(16±2)个、(42±3)个(F=73.446,P<0.001),两观察组与健康组差异明显,而两观察组无明显差异。qRT-PCR和Western blot结果显示,两观察组中Cofilin蛋白表达明显高于健康组。结论 COPD患者肺泡及外周血单核源性巨噬细胞吞噬烟曲霉功能降低可能与Cofilin蛋白升高有关。
