miR-15a-5p对胃癌细胞增殖的影响

摘要
目的观察miR-15a-5p对胃癌细胞增殖的影响。方法将miR-15a-5p mimics、mimics control、miR-15a-5p inhibitor、inhibitor control分别转染至胃癌SGC-7901和BGC-823细胞,应用实时定量PCR分别检测miR-15a-5p的表达水平以验证miR-15a-5p mimics对miR-15a-5p表达的影响及miR-15a-5p inhibitor对miR-15a-5p表达的影响。CCK-8实验检测miR-15a-5p mimics及miR-15a-5p inhibitor对胃癌SGC-7901和BGC-823细胞增殖的影响。结果实时定量PCR表明与转染mimics control相比,转染miR-15a-5p mimics后SGC-7901细胞和BGC-823细胞中miR-15a-5p的表达水平显著升高(P <0. 05)。与转染inhibitor control相比,转染miR-15a-5p inhibitor后SGC-7901和BGC-823细胞中miR-15a-5p的表达有下降趋势,但差异无统计学意义(P> 0. 05)。CCK-8实验表明过表达miR-15a-5p可抑制胃癌细胞SGC-7901和BGC-823的增殖(P <0. 05),miR-15a-5p inhibitor促进SGC-7901和BGC-823细胞增殖(P <0. 05)。结论 miR-15a-5p可能抑制胃癌细胞的增殖。
