miR-125b-5p靶向STAT3调节JAK抑制骨髓增生异常综合征细胞增殖

摘要
目的 探究miR-125b-5p对骨髓增生异常综合征细胞增殖的作用及可能机制。方法 qRTPCR检测骨髓增生异常综合征患者骨髓组织miR-125b-5p表达;将miR-125b-5p模拟物转染骨髓增生异常综合征SKM-1细胞,CCK-8和Edu实验检测细胞增殖;qRT-PCR检测STAT3、JAK1和JAK2 mRNA表达;Western blot检测STAT3、JAK1和JAK2蛋白表达;生物信息学预测STAT3 3'非翻译区miR-125b-5p潜在结合位点,并通过双荧光素酶报告基因实验验证。结果 miR-125b-5p在骨髓增生异常综合征患者骨髓组织中表达上调。上调miR-125b-5p抑制SKM-1细胞增殖,抑制SKM-1细胞STAT3、JAK1和JAK2表达。miR-125b-5p靶向结合STAT3。结论miR-125b-5p通过靶向抑制STAT3表达调节JAK抑制骨髓增生异常综合征细胞增殖。
