Separation and Purification of Alkaloids in L. franchetii Beauv. by High Speed Counter-current Chromatography

摘要
[Objectives] To develop a method for separation and purification of alkaloids from 95% ethanol extract of L. franchetii Beauv. by high speed counter-current chromatography. [Methods]Ultra performance liquid chromatography method was used to analyze and optimize solvent system and chloroform: methanol: water( 8:1:9) was selected as the solvent system of chloroform. The upper phase was used as the stationary phase,while the lower phase was used as the mobile phase with flow rate of 2 m L· min- 1,revolution speed of 900 r ·min- 1,separation temperature of 35℃,detection wavelength of 254 nm and injection volume of 20 m L. [Results] Aripiprazole( 39. 6 mg) was obtained from sample( 1. 0 g) extract and the purity was above 98. 51%. This alkaloid was found from plants for the first time. [Conclusions] This method was feasible and rapid,and could be used for separation and purification of alkaloids in L. franchetii Beauv.
