铁蓄积通过干扰Wnt信号通路导致骨质疏松

摘要
取6个月龄的野生型小鼠和6个月龄的铁调素基因敲除(Hepcidin-KO)小鼠(铁蓄积模型)的股骨标本, 进行Micro-CT检测, 以Western印迹和免疫共沉淀检测Wnt信号通路中相关指标的变化。与野生型小鼠相比, Hepcidin-KO小鼠骨量明显下降, 骨组织内β-连环蛋白(β-catenin)、TCF4/TCF7L2、FOXO3a蛋白表达无明显变化, 而β-catenin蛋白与FOXO3a结合增多, 与经典Wnt通路中TCF4/TCF7L2的结合减少, 提示铁蓄积可能通过影响经典的Wnt/β-catenin信号通路影响骨形成, 进而导致骨质疏松。
