[Objective]A submerged fermentation medium of Pleurotus tuber-regium was developed for efficient production of polysaccharide and biomass. [Methods] By employing single factor experiment and orthogonal design,the effects of carbohydrate and nitrogen sources and cations on the Pleurotus tuber-regium fermentation were investigated with the intracellular and extracellular polysaccharides as indices. [Results]Glucose was found to be the optimal carbohydrate source,favorable for cell growth,metabolite production of extracellular polysaccharide( EPS) and intracellular polysaccharide( IPS),the yield of polysaccharides per unit biomass. Yeast extract as nitrogen sources was suitable for the cell growth,mycelial biomass and the yield of extracellular polysaccharide. The type of cations source also affects polysaccharide production,and EDTA performed best in promoting cell growth and metabolites accumulation. Under the optimized conditions,the production of biomass,intracellular polysaccharide,extracellular polysaccharide,the yield of total polysaccharide and dried cell biomass was 3. 34 g / L,1. 92 g / L, 80. 34 mg / g,8. 13% and 23. 62 g / L,respectively. [Conclusions]The optimized conditions for submerged fermentation of Pleurotus tuber-regium provided scientific basis for the cultivation of Pleurotus tuber-regium.
