miR-29a基于BMP2/Runx2对肋骨骨折大鼠的干预作用

摘要
目的:探讨miR-29a调控骨形态发生蛋白2(BMP2)/矮小相关转录因子2 (Runx2)在大鼠肋骨骨折愈合中的作用。方法:建立SD大鼠肋骨骨折模型,分为miR-29组和对照组,每组20只。14 d和28 d各处死动物10只,行X线和micro-CT检查,观察骨折愈合情况,分别采用实时荧光定量PCR(RT-qPCR)法和western blotting法检测Runx2和BMP2 mRNA和蛋白表达水平。结果:X线摄片显示自14 d开始两组均有骨痂开始形成,至28 d骨痂塑形改建,骨折愈合。与对照组相比,miR-29组骨密度、骨矿物质含量及骨骼体积比均显著增加,Runx2和BMP2 mRNA和蛋白表达量均明显升高(P<0.05),且28 d时Runx2和BMP2表达量较14 d时明显下降(P<0.05)。结论:miR-29a能够通过Runx2/BMP2信号通路促进骨折部位骨小梁较早的形成和改建,促进矿物质沉积,从而加速骨折愈合。
