桑叶中1-脱氧野尻霉素的纯化及含量测定

摘要：

目的提取纯化桑叶中1-脱氧野尻霉素(DNJ),并用柱前衍生化高效液相法进行含量测定。方法桑叶加0.05mol·L-1盐酸溶液超声提取,提取物进行反相柱色谱纯化,纯化物用柱前衍生化-高效液相-荧光检测法测定DNJ的含量。结果桑叶中DNJ的含量为0.27%～0.49%,纯化物中DNJ的含量达到了10.7%,DNJ在0.6～60μg·mL-1内浓度与峰面积呈良好的线性关系(r=0.9997),平均回收率为104.46%。结论运用反相色谱技术分离纯化DNJ简单易行,含量测定方法精确,重复性好。
