The effect of cytoplasmic regions of LIF receptor on activating MAPK p42/44in HL-60 cell

摘要
Objective:To studytherelationbetweenthecytoplasmicregionof eachsubunitof leukemiainhibitoryfactor(LIF)receptorandmitogen-activatedproteinkinase(MAPK)in humanleukemialineHL-60for studyingthe mechanism of leukemiacellproliferationdisorder.Methods:Wepreparedthechimericalreceptorwithexchangedcytoplasmicdo-main(190/130,130/190),expressediton themembraneof HL-60,andthenboundLIFwiththewildtypereceptorcompet-itively.Thecytoplasmicregionhomodimerization(190cyt-190cyt,130cyt-130cyt)initiatedintracellularsignaltransduc-tion,cellproliferationanddifferentiation.MAPKphosphorylationlevelwasobservedby meansof immunoblottingand immunobiochemistry.Results:Higherlevelof MAPKp42/p44phosphorylation(Thr202Tyr204)andfastercellproliferation weredeterminedingroup190/130comparedwiththewildtypereceptor.Thoseingroup130/190werelowerthantheoth-ers.Conclusion:Thecytoplasmicdomainof gp130involvesin theinductionof MAPKactivationandthecellprolifera-tionof HL-60.
