高迁移率族蛋白B1在卵巢癌组织及单克隆细胞亚系中的表达及其临床意义

摘要：
目的探讨高迁移率族蛋白B1(HMGB1)在卵巢癌组织及单克隆细胞亚系中的表达及其临床意义。方法采用免疫组化法检测80例卵巢上皮性肿瘤(良性24例、恶性56例),及20例正常卵巢组织中HMGB1的表达;采用Agilent芯片检测高转移单克隆细胞亚系S1和低转移单克隆细胞亚系S21的基因表达差异,同时采用细胞爬片、Real time RT-PCR、Western blot等方法检测高转移单克隆细胞亚系S1、A1,以及低转移单克隆细胞亚系S21、A14中HMGB1的表达。结果 HMGB1在卵巢癌中的表达明显高于良性卵巢肿瘤和正常卵巢组织(P<0.01),且与卵巢癌的临床分期、分化程度及有无淋巴结转移密切相关(P<0.05)。HMGB1在高转移单克隆细胞亚系中高表达,在低转移单克隆细胞亚系中低表达,与细胞的侵袭转移密切相关(P<0.05)。结论 HMGB1的高表达与卵巢肿瘤组织分化程度低、高度黏附和侵袭生长等恶性表型密切相关,可能对其恶性表型起正性调节作用。
