摘要
Objective. Findingmethods that can interfere withWnt/beta-cateninsignalinghas becomeanimportant research direction in inhibiting coloncancermetastasis. Mesoporous silica nanoparticles canefficientlycarry and releasedrugs.Therefore, combiningligustrazine,miR-570,and mesoporous silicananoparticlesas carriers will provide a theoretical basisfor developmentof new therapeutic strategiesanddrugs.Methods. We herein prepared mesoporous silica-loadedligustrazinenanoparticlesand usedthemto culture HT-29 cells;we observed biologicalbehaviorof HT-29 and explored the levels of miR-570and Wnt2/beta-catenin.Results. Mesoporous silicananoparticlesloadedwith Ligustrazine were successfullyprepared. Ligustrazineinhibitedmetastasis of HT-29 cells.Mesoporous silicananoparticles carrying li-gustrazineincreasedthe expressionof miR-570and reduced Wnt/beta-cateninin HT-29 cells.Moreover,overexpressionof miR-570 inhibitedHT- 29 cancercell metastasis and Wnt/beta-catenininhibitionled toinhibition of HT-29 cell metastasis, whileinhibiting miR-570expressionreversed the effect of mesoporoussilicananoparticlescarrying ligustrazine,thereby accelerating HT-29 cell metastasis.Conclusion. miR-570can inhibit Wnt/beta-cateninexpression.Mesoporous silicananoparticles carrying ligustrazinecan promotemiR-570to inhibitWnt/beta-cateninexpression,leadingto inhibition of HT029cell metastasis
