Preparation, structure characterization and solubility of one cocrystal and one salt of Imatinib with Citric acid and Fumaric acid

摘要
One cocrystal of Imatinib with citric acid (S1) and one salt with fumaric acid (S2) have been prepared and investigated systematically. Crystal structure analysis reveals that reliable O-H center dot center dot center dot O, O-H center dot center dot center dot N, N-H center dot center dot center dot N and N-H center dot center dot center dot O hydrogen-bonding interactions exist in both co-formations. Single crystal and powder X-ray diffraction, Fourier transform infrared and differential scanning calorimetry were used to characterize the two crystals. The solubility of the solid were also evaluated, and two co-formations (S1, S2) had a more than 100 times of solubility and a faster dissolution rate compared to the pure drug in pH 6.8 buffer.
