Optimization of fermentation medium and conditions for inosine production using response surface methodology

摘要
To optimize the medium and conditions for fermentation culture of inosine by response surface analysis. The important parameters were screened by Plackeet-Burman design from 9 factors among inosine shake flask fermentation process and the results showed that inosine yield could be significantly affected by the concentration of glucose, concentration of coin liquid, liquid medium volume and inoculation amount. The path of steepest ascent was used to approach to the biggest region of inosine yield, and the optimum values of the parameters were obtained by Box-Behnken design were as follows: glucose concentration 236.87 g/L, corn liquid concentration 23.91 g/L, liquid medium volume 7 mL and inoculation amount 6.5%. Under such optimum process conditions, the inosine yield reached 8.26 g/L which increased 49.9% than that under the initial conditions.
