Direct electrochemistry of hemoglobin with 1-Ethyl-3-methylimidazolium ethylsulfate on carbon ionic liquid electrode and its biosensing

In this paper a carbon ionic liquid electrode (CILE) was prepared with an ionic liquid (IL) 1-ethyl-3-methylimidazolium ethylsulfate ([EMIM]EtOSO3) as the modifier. Then hemoglobin (Hb) in pH 8.5 phosphate buffer solution and [EMIM]EtOSO3 were assembled alternately on CILE to get a {IL/Hb}n multilayer film modified electrode. Electrochemical experiments indicated that a pair of well-defined quasi-reversible redox peaks appeared with the formal peak potential (E0′) as-0.219 V (vs. SCE), which was the characteristic of Hb heme Fe(III)/Fe(II) redox couples. Electrochemical behaviors of Hb in the multilayer modified electrode were investigated and the results exhibited that the presence of IL in the film showed an obvious promotion to the direct electron transfer of Hb. The {IL/Hb}n/CILE showed good electrocatalytic ability to the reduction of trichloroacetic acid, which provided a new platform to fabricate the third-generation biosensor based on the direct electrochemistry of redox protein with the presence of IL.
