Enrichment of metal chelating peptides from wheat germ protein by immobilized-metal ion affinity chromatography

Wheat germ protein has been hydrolyzed by three proteases (Alcalase 2.4L, Flavourzyme 500MG and Papain) respectively. The hydrolysates with highest metal chelating ability have been injected into immobilized-metal ion affinity chromatography to enrich metal chelating peptides. The relative molecular mass distribution and amino acid composition of the metal chelating peptides were analyzed. The results showed that the hydrolysates could be obtained on condition as Alcalase 2.4L and 200 min, and the highest metal chelating ability reached (69.62±0.96)%. The relative molecular mass of metal chelating peptides separated from wheat germ protein hydrolysates was concentrated in the range of 180~2 000. Besides, the metal chelating peptides had high content of Glu and Asp with the level of (22.64±1.50)% and (14.93±0.24)% respectively. ©, 2015, Editorial Department, Chinese Cereals and Oils Association.
