Analysis of spiral reentry generation mechanisms with cardiac intracellular Ca2+ pocket and temporal Ca2+ burst phenomenon

Ventricular tachyarrhythmia or ventricular fibrillation. However it was still unknown about Ca2+ mechanisms on the condition of 2D/3D cardiac tissue structure. We developed Vm/Ca2+ dual mapping and precisely registration two signals systems as previously reported, and measured and analyzed Vm/ Ca2+ dynamics during initiated VT to VF changes. Langendorff perfused rabbit heart was stained with Rhod-2 and Rh237 simultaneously. Vm and Ca2+ signal were split by dichroic mirror and recorded as 512 * 512 pixels, 250 fps by two high speed cameras. VT were initiated by cross-field stimulation with S1-S2 protocol. In the episodes of transition VT to VF, Ca2+ sinkhole like pocket area and extremely increased Ca2+ signal (Ca2+ burst) were confirmed and then wave breakup on membrane potential signals and spiral reentry were generated. © 2014, Japan Soc. of Med.
