Inhibitory effect of fermented wheat germ extract on human colon carcinoma cell line HT-29 xenograft in nude mice

To study the effect of fermented wheat germ extract with Lactobacillus plantarum dy-1 isolated from pickles on tumor growth inhibition of human colon carcinoma cell strain HT-29 xenograft in BALB/c nude mice, and explore the possible mechanisms. HT-29 cells were injected subcutaneously into nude mice about 4-6 weeks to establish model of transplanted tumor (inoculum size: 1×106~2×107 cells/mice) to establish human colon carcinoma cell line HT-29 xenograft in nude mice model, model nude mice were divided randomly into five groups treated with gavage, viz the model group given model group(15 mL/kg/d, Physiological saline), 5-FU group [inject 25 mg/(kg·d), 7d], mixture group [2 g/(kg·d) LFWGE and 25 mg/(kg·d) 5-FU, 7d], high-dose group [2 g/(kg·d) LFWGE] and low-dose group [1 g/(kg·d) LFWGE]. The inhibitory effect of fermented wheat germ extract on the growth of tumors in nude mice was observed and the inhibitory rate was calculated. Finally we determined the levels of serum lipid profiles, liver antioxidant capacity, Immune factors and others. The inhibitory of the 5-FU group, mixture group, high-dose group, low-dose group were 30.08%, 47.80%, 41.72%, 40.70% mm3, respectively. LFWGE was non-toxic side effects on the liver and spleen. Relative to model group, LFWGE was able to reduce MDA content and increase SOD, CAT, T-AOC, GSH-PX active. LFWGE also can increase the expression of IFN-γ, IL-4, TNF-α. LFWGE has the inhibitory effects on the human colon carcinoma cell strain HT-29 xenograft in BALB/c nude mice. The mechanism could possibly contribute to increase the expression of immune factors, reduce the damage of liver and spleen, increase liver antioxidant capacity and others. © 2016, Chinese Institute of Food Science and Technology.
