Polysaccharides existing in advanced plant, animal and microorganism are one of very important natural macromolecules. Many bioactive polysaccharides have good effects on immune modulation, anti-tumor, anti-oxidant and gut bacteria regulation bioactivities. The clearly elucidating in polysaccharide structure would guide deeply understanding their bioactivities and be beneficial for the basic structure research and applications. In this paper, it is briefly introduced about the isolation, purification and structural elucidation strategies of polysaccharides from natural resources. Research progress is highly emphasized on structural characterization of glucan, glucomannan, xyloglucan, arabinoxylan and pectin. We also propose some problems in the present research and point out our own view. We suggest that the combined utilization of partial degradation, methylation and NMR could be taken as the key method for polysaccharide structural elucidation, by reviewing lots of research papers and summarizing our own experiences. Meanwhile, other methods such as ESI-MS, Maldi-Tof-MS and ion mobility-mass spectrometry could also be used additionally to establish a advanced and systematic method for fine structure characterization of polysaccharides. © 2017, Editorial Office of Journal of CIFST.
