The departure strains of Bacillus cereus are handling by three kinds of mutagenesis, and get a phospholipase C high-yield strains of B.cereus-3. Selection of the precipitation ring diameter and the number of viable counts as a reference. By response surface to optimize the fermentation conditions of the strain B. cereus-3, the result for that: the inoculum size is 2.5%, speed is 160 r/min, fermentation temperature is 30.5℃ and the fermentation time is 18.5 h. At the time, the precipitation ring diameter is maximum, and phospholipase C activity is 26.3 U/mL. The crude enzyme solution were isolated and purified, and used SDS-PAGE method to measure the molecular weight of the purified phospholipase C, molecular weight between about 29-34 ku. © 2018, Editorial Office of Journal of CIFST.
