Synthesis of low degree of substitution sulfobutyl ether-β-cyclodextrin and their structural characterization

A novel process for producing sulfobutyl ether-β-cyclodextrins with low substitution degrees was developed using the different pH of alkaline electrolyzed water as medium by microwave heating. Sulfobutyl ether-β-cyclodextrins with average substitution degrees of 2-3 were synthesized by changing the molar ratio of β-cyclodextrin to 1,4-butane sultone, the volume of alkaline electrolyzed water with different pH and the heating power at 75 ℃. The product yields were 75.2%, 73.5% and 70.1% respectively. The effects of pH of alkaline electrolyzed water on product yields were also investigated. The structure of products was characterized by NMR, IR, MS, Elemental analysis and its substitution degrees were calculated. The results show that sulfobutyl ether-β-cyclodextrins with low substitution degree can be synthesized through the novel synthesis process, and the highest yields can be obtained when the pH of alkaline electrolyzed water is 12.7. In addition, the average substitution degrees calculated by element analysis are more reliable than those obtained by 1H NMR and MS. © 2020, Editorial Board of Journal of Chemical Engineering of Chinese Universities".
