The structural parameters affecting the efficiency of separation and purification of chromatography media are introduced. Several common methods to determine pore structure of chromatographic media, such as mercury intrusion porosimetry, nitrogen adsorption method, small angle X-ray scattering, electron microscopies (TEM and SEM), and inverse size-exclusion chromatography (ISEC) are summarized. The basic principle of ISEC and its involved probe types are described. The related theories of ISEC, especially solute radius, chromatographic theory and size-exclusion chromatography (SEC) random theory are discussed. The advantages and disadvantages of ISEC are compared. The application of ISEC in evaluating pore size structure of chromatographic media is introduced.
