Growth inhibition of interleukin-2 receptor gene-transduced peripheral blood lymphocytes on human ovarian cancer cells

摘要
目的 为研究IL 2R基因转染PBL对人卵巢癌细胞生长抑制的影响。方法 用脂质体Fugene作载体将IL 2和IL 2R基因分别转染入人卵巢癌细胞株 3AO和人外周血白细胞 (PBLs) ,转染 2 4小时后分别将转染和非转染的PBLs与转染和非转染的 3AO细胞混合培养 4 8小时 ,并用MTT法测定PBLs对 3AO细胞的杀伤作用。结果 转染前后 3AO细胞和PBLs细胞形态无明显变化。 3AO能转染IL 2基因并表达IL 2mRNA ,PBLs能转染IL 2R基因并表达IL 2RmRNA。IL 2基因修饰 3AO细胞可增强淋巴细胞对 3AO细胞的抑制作用 ,同时联合IL 2R基因转染PBLs和IL 2基因修饰 3AO细胞可显著增强其淋巴细胞对 3AO细胞的抑制作用。结论 IL 2R基因转染效应细胞可增加IL 2基因转染肿瘤细胞的抗肿瘤免疫效应 ,这一方法为卵巢癌IL 2基因治疗开辟了新的治疗途径
