Hsa-let-7b-5p抑制非小细胞肺癌的增殖、迁移和侵袭

摘要
目的 探讨hsa-let-7b-5p在非小细胞肺癌临床诊断和靶向治疗中的价值。方法 收集2020年9月-10月的13对哈尔滨医科大学附属肿瘤医院病理类型相同的非小细胞肺癌癌组织及其癌旁组织,通过RNA-Seq获得差异表达的miRNA,并通过qRT-PCR验证候选miRNA在非小细胞肺癌组织和细胞中的表达水平。通过CCK-8法、克隆形成实验、划痕实验和Transwell实验验证miRNA对非小细胞肺癌增殖、迁移和侵袭能力的影响。利用MiRDB、TarBase和ENCORI数据库预测靶基因,并利用FunRich工具进行基因富集分析。结果 与癌旁组织及正常支气管上皮细胞系(Human bronchial epithelial cell, HBE)相比,hsa-let-7b-5p在非小细胞肺癌组织及非小细胞肺癌细胞系(A549、H1299、H358和H460)中低表达(P<0.05)。hsa-let-7b-5p的高表达可以抑制非小细胞肺癌增殖、迁移和侵袭能力(P<0.01)。靶基因预测及富集分析结果显示hsa-let-7b-5p的378个靶基因及其相关基因中,147个基因与hsa-let-7b-5p的表达呈负相关(P<0.05)。结论 Hsa-let-7b-5p对非小细胞肺癌的增殖、迁移和侵袭有抑制作用。
