DETECTION OF GUANYL CYCLASE IN MAMMALIAN TISSUES

Guanyl cyclase was assayed by determining the rate of conversion of [α-32P]GTP to cyclic 3′,5′-[32P]GMP. A two-step chromatographic procedure was developed for isolating the product. The enzyme was found in the soluble fraction after homogenizing tissues in 0.3 M sucrose. The reaction showed a pH optimum near 7.6 and was dependent on Mn2+. The relative enzymatic activity for tissues of the rat was lung >> brain = spleen > kidney = liver = heart. © 1969.
